Pharmacodynamic Parameters of Sildenafil

The basic Sildenafil citrate metabolite is N-desmethyl. The fact is that N-desmethyl metabolite circulates in the blood plasma, just as Sildenafil citrate does. About 20% of pharmacological effect of Sildenafil citrate is provided namely by N-desmethyl metabolite.

After oral intake, Sildenafil citrate is excreted from the body in form of active and inactive metabolites.

  • About 80% of the dose of Sildenafil citrate is excreted with feces.
  • About 13% of Sildenafil citrate dose is excreted with urine.

It must be said that the excretion rate of Sildenafil citrate (and its metabolites) from the body decreases in elderly men over 65 years old with hepatic disorders and severe kidney disease. Hence, this group of patients might need an individual dose adjustment of Sildenafil citrate.

Pharmacodynamic features of Sildenafil citrate

Clinical trials, which involved men with psychogenic or organic erectile dysfunction, demonstrate that:

  • Erection recovery takes place approximately in 60 minutes after the use of a single dose of Sildenafil citrate.
  • The duration of Sildenafil citrate pharmacological effect may reach 4 hours.
  • Within 1-3 hours after the intake of Sildenafil citrate, men’s blood pressure may drop down.
  • An increase of Sildenafil citrate daily dose helps to improve the efficiency of erectile dysfunction treatment.
  • Maximal daily doses of Sildenafil citrate never change an intraocular pressure and never affect the vision acuity in men.
